The user.css must be place inside the directory
media/templates/site/cassiopeia/css

Remember to clear cache also

I did what you suggested and put the following code to change the header color and nothing happened.

--cassiopeia-color-primary: #2F9C95;
--cassiopeia-color-hover: #174F4B;
--cassiopeia-color-link: #1C5E5A;
}
